In the present study, we evaluated the analgesic effect of sildenafil citrate and sildenafil in the presence of COX-2 inhibitors. COX-2 inhibitors reduce the prostaglandin-dependent COX-2 signaling in the corpus cavernosum. The inhibitory effect of sildenafil citrate was evaluated using a COX-2 inhibitor assay. The results showed that sildenafil citrate reduced the prostaglandin-dependent COX-2 signaling in the corpus cavernosum, while sildenafil did not affect the prostaglandin-dependent COX-2 signaling in the corpus cavernosum. The inhibition of COX-2 by sildenafil can be a useful treatment option for patients suffering from PAH. Copyright © 2022. Published by Elsevier Inc. All rights reserved.

Introduction

Sildenafil citrate (Viagra), a PDE-5 inhibitor, is a selective PDE-5 inhibitor that works by inhibiting the phosphodiesterase type 5 (PDE5) in the corpus cavernosum. Sildenafil citrate is widely used to treat erectile dysfunction (ED) and pulmonary hypertension (PH). However, the clinical use of sildenafil citrate is limited by the low bioavailability and limited effectiveness of the drug, which may cause adverse effects and drug resistance. In order to achieve a higher potency of sildenafil, sildenafil is also widely used as an alternative to other PDE5 inhibitors, such as tadalafil, vardenafil, and riluzole. However, it has not been studied the interaction between these PDE-5 inhibitors and sildenafil. In this study, we explored the potential interaction between sildenafil and sildenafil in the presence of COX-2 inhibitors in the treatment of patients with PAH. The aim of this study was to investigate the effect of sildenafil citrate and sildenafil in the presence of COX-2 inhibitors and sildenafil citrate on the prostaglandin-dependent COX-2 signaling in the corpus cavernosum. The study was conducted in accordance with the Declaration of Helsinki and Good Clinical Practice, and approved by the Ethics Committee of the Shanghai Jiaotong Hospital, Shanghai, China. A written informed consent was obtained from all participants before the study. This study was carried out in accordance with the approved guidelines of the Shanghai Jiaotong Hospital and the Ethics Committee of the Shanghai Medical University, Shanghai, China. All participants gave written informed consent prior to the study.
